Which bus station you depart from depends on where you are staying.
If you are in Georgetown, by far the easiest place to take a bus from is under the KOMTAR building.
However, there are now only a few departures a day from here.
Another good option (with many more frequent departures) is to take the ferry over to Butterworth and get a bus from Penang Sentral / Butterworth Bus Terminal (adjoining the ferry jetty).
Getting to Sungai Nibong Bus Terminal is a bit more hassle if you are staying in Georgetown (you will have to take a RapidPenang bus or taxi).

You can purchase bus tickets from many different places around Penang Island.
Most hotels and guesthouses will sell you tickets, as well as all the travel agents around the Chulia Street area in Georgetown.

If you want to travel by train to Melaka from Penang, you will have to head over to the mainland and take a train to the nearest railway station to Melaka at Tampin (also called Pulau Sebang).
To get to Tampin by train you can either:
1) take an ETS train all the way from Butterworth to Tampin (only limited services).
2) take a more frequent train from Butterworth to KL Sentral, then change on to a KTM Komuter train from KL to Tampin.
Once you arrive at Tampin Railway Station, you are still 40 km away from Melaka.
To get to Melaka from Tampin you can take a taxi or walk into town a catch a cheap local bus.
Another option you may want to consider is to travel from Penang to TBS Bus Terminal by train, then take a bus from TBS to Melaka Sentral.
TBS Bus Terminal in KL is located right next to the Bandar Tasik Selatan Train Station.
You can take a train directly from Butterworth (limited services) or take a train to KL Sentral, transfer to a KTM Komuter train to TBS, then take a bus from TBS to Melaka Sentral.
Malindo Air operates flights from Penang International Airport to Melaka Airport, four times a week.
You may also want to look at flying from PEN to KUL.
From KUL you can then take a regular bus to Melaka.

This is a long 500 km drive along the North-South Expressway (E1), so you should consider stopping off along the way to break the journey.
If you want to do this car journey in one go it should take around 7 hours.
